Description: INTRODUCTION AND AIMS: AQP2 is expressed in the apical plasma membrane and subapical vesicles in the collecting duct principal cells. Collecting duct water reabsorption in response to vasopressin is mediated by translocation of intracellular AQP2-bearing vesicles to the apical plasma membrane and long-term regulation of the AQP2 expression. We recently demonstrated that luminar fluid shear stress exerts an important effect on the AQP2 trafficking to the apical plasma membrane of primary cultured inner medullary collecting duct (IMCD) cells (Jang et al, Integr Biol, 2011). In this study we aimed to examine the effects of altered luminal microenvironment, e.g., transepithelial osmotic gradient or pH gradient on dDAVP-induced AQP2 trafficking in the IMCD cells of rat kidney. METHODS: IMCD cells were seeded on transwell system with hyperosmolar culture medium (640 mOsm/KgH 2 O, pH 7.4, both at the apical and basolateral side) for 5 days, and then these cells were exposed to transepithelial osmotic gradient for 30 min (340 mOsm/KgH 2 O at the apical side vs. 640 mOsm/KgH 2 O at the basolateral side) or pH gradient for 30 min (pH 6.4 at the apical side vs. pH 7.4 at the basolateral side) in the absence or the presence of dDAVP stimulation (10-9M for 30 min) applied to the basolateral side. RESULTS: Laser scanning confocal microscopy (z-sectioned images) revealed that 30 min exposure to culture medium with either lower osmolality (340 mOsm/KgH 2 O) or lower pH (pH 6.4) at the apical side substantially reduced the dDAVP-induced AQP2 trafficking. Moreover, cell surface biotinylation assay demonstrated significantly decreased expression of dDAVP-induced phosphorylated AQP2 (Ser 256) in the cells exposed to either lower osmolality (340 mOsm/KgH 2 O) or lower pH (pH 6.4) at the apical side. CONCLUSIONS: Taken together, altered luminal microenvironment per se , e.g., osmolality and pH, could exert a profound effect on dDAVP-induced AQP2 trafficking in the collecting duct principal cells.
